Shanghai experts confirmed - streptococcal DNA can cause psoriasis

A study led by Dr. Zheng Jie from Shanghai Jiejin Hospital has provided new insights into the role of streptococcal DNA in triggering psoriasis. The research, titled "Bacterial DNA components in streptococcal antigens enhance the proliferation of peripheral blood mononuclear cells (PBMC) and activation in patients with psoriasis vulgaris," was published online on July 16, 2009, in the Journal of Investigative Dermatology. The study focused on how streptococcal DNA influences immune responses in psoriasis patients. Researchers observed that PBMCs from these patients showed a stronger proliferative response when exposed to streptococcal antigens (SA) compared to those from healthy individuals. To further investigate, they used DNase-I to remove nucleic acid components from SA, creating a modified version called non-NASA. When PBMCs were stimulated with non-NASA, their proliferative response was significantly reduced compared to SA-stimulated cells. Additionally, the study found that non-NASA led to lower levels of CD69 expression—particularly on T cells associated with skin homing—and reduced secretion of interferon-alpha (IFN-α). However, when synthetic CpG-A, a DNA mimic of streptococcus, was added during stimulation, the effects of SA were restored. This suggests that the DNA component plays a crucial role in activating immune cells involved in psoriasis. Interestingly, the expression of CD86 on B cells and the secretion of IFN-γ and TNF-α remained largely unchanged between groups. But when CpG-B was introduced alongside the stimulation, CD86 expression increased significantly. These findings highlight the importance of streptococcal DNA in promoting immune activation, particularly in T cells, which may contribute to the progression of psoriasis. This research underscores the potential link between bacterial DNA and autoimmune skin conditions, offering new directions for understanding and treating psoriasis. Meditech Medical Network
